The low-dose influenza vaccine (GSK1247446A) is an investigational trivalent inactivated split-virion vaccine developed by GSK Biologicals for the prevention of seasonal influenza in adults aged 60 years and older. It contains a reduced dose of hemagglutinin (HA) antigens from three influenza strains, formulated with GSK's proprietary AS03 adjuvant system—an oil-in-water emulsion containing squalene, alpha-tocopherol, and polysorbate-80. By utilizing the adjuvant to compensate for the lower antigen content, the vaccine aims to elicit protective haemagglutination-inhibition (HI) antibody titers comparable to or better than standard-dose vaccines while potentially improving the efficiency of vaccine production. It has been evaluated in Phase II clinical trials to assess its immunogenicity and safety profile.
