Overview

LOXHD1 TCR T cells is a preclinical adoptive cell therapy (ACT) consisting of autologous CD8+ T cells engineered to express a high-avidity αβ T cell receptor (TCR) targeting Lipoxygenase Homology Domains 1 (LOXHD1). LOXHD1 is a tumor-associated antigen (TAA) whose expression in Ewing Sarcoma (EwS) is driven by the pathognomonic EWSR1::FLI1 oncofusion. The therapy is designed for HLA-A*02:01-positive patients and targets LOXHD1 peptide epitopes presented on MHC class I molecules. In preclinical studies, these engineered T cells demonstrated cytotoxic activity against EwS tumor cell lines and led to tumor eradication in mouse xenograft models. LOXHD1 expression is otherwise highly restricted to the inner hair cells of the cochlea and adult testis, making it a relatively tumor-specific immunotherapeutic target for pediatric bone and soft tissue malignancies.
