LP-005 is a novel bifunctional complement antibody fusion protein designed to inhibit both complement component C3 and C5. This dual-targeting approach provides broad inhibition of the complement cascade, with demonstrated superior bioactivity in classical (CP), alternative (AP), lectin (LP) pathways, and C3b deposition assays compared to existing therapies such as eculizumab, ravulizumab, crovalimab, pozelimab, and narsoplimab. Engineered for optimized surface charge and FcRn binding, LP-005 is intended as a first-in-class Q4W (once every four weeks) bi-functional complement inhibitor. It is being developed primarily for the treatment of paroxysmal nocturnal hemoglobinuria (PNH) and IgA nephropathy (IgAN), with additional development planned in neurology and ophthalmology indications. Preclinical studies have shown favorable pharmacokinetic/pharmacodynamic profiles and safety data[1][3][6].
