LP-284 is a novel small molecule, next-generation acylfulvene that acts as a DNA-damaging agent with a synthetically lethal mechanism of action. It preferentially targets and kills cancer cells—especially those with deficiencies in DNA damage repair pathways such as ATM or ATR mutations—by inducing lethal DNA double-strand breaks. LP-284 has demonstrated potent anti-tumor activity in preclinical models of B-cell non-Hodgkin lymphoma (NHL), including high-grade B-cell lymphoma (HGBL), mantle cell lymphoma (MCL), and diffuse large B-cell lymphoma (DLBCL). The drug is particularly effective against tumors resistant to standard therapies like ibrutinib and bortezomib. Mechanistically, it also functions as a potent depleter of CD19+ and CD20+ B cells, making it relevant for both hematologic malignancies and potentially autoimmune disorders such as lupus nephritis. Developed using AI-driven approaches by Lantern Pharma, LP-284 is currently being evaluated in Phase 1 clinical trials for relapsed/refractory lymphomas and select solid tumors[1][3][4][5][6][7][8][9].
