This combination is a three-drug regimen consisting of **LP002**, a recombinant humanized monoclonal antibody against programmed death ligand-1 (PD-L1), plus **cisplatin**, a platinum-based chemotherapy agent, and **fluorouracil (5FU)**, a pyrimidine analog antineoplastic agent. LP002 binds PD-L1, blocking its interaction with PD-1/B7.1, thus enhancing antitumor immune responses. Cisplatin forms DNA crosslinks, inhibiting DNA synthesis and function, leading to cell death. Fluorouracil, after metabolic activation, inhibits thymidylate synthase and gets incorporated into RNA and DNA, disrupting cell division and function. The combination is under investigation primarily for resectable, PD-L1 positive, locally advanced gastric and gastroesophageal junction (GEJ) cancers as perioperative therapy.
