LP10 + OVA8 + CpG + LLP2A is a programmable peptide nucleic acid (PNA)-based nanovaccine (PVN) designed for targeted cancer immunotherapy. The platform utilizes an 11-mer PNA scaffold (LP10) to facilitate the precise assembly of three functional components: a PNA-modified ovalbumin antigenic peptide (OVA8, containing the SIINFEKL sequence), a CpG oligodeoxynucleotide adjuvant, and a PNA-derivatized LLP2A ligand. The LLP2A component specifically targets the activated α4β1 integrin (also known as VLA-4) expressed on the surface of immune cells and certain tumor cells, such as melanoma. This targeted approach enhances the delivery of antigens and adjuvants to dendritic cells, thereby boosting antigen presentation and eliciting potent CD8+ T cell and natural killer (NK) cell immune responses. Preclinical studies in B16-OVA syngeneic melanoma models have demonstrated that this nanovaccine can induce significant tumor regression and prolong survival.
