LP11W is an apoptogenic oncolytic adenovirus type 5 (HAdV5) vector developed for the treatment of solid tumors, specifically head and neck squamous cell carcinoma (HNSCC). The vector is genetically engineered to be defective in the expression of the E1B-19K protein, a viral homolog of the anti-apoptotic Bcl-2 protein that normally prevents premature host cell death during viral replication. By lacking E1B-19K, LP11W induces enhanced apoptotic and cytotoxic responses in infected tumor cells. Preclinical evaluation in immunocompetent Syrian hamster models has demonstrated that intratumoral administration of LP11W significantly suppresses the growth of both subcutaneous and orthotopic cheek pouch tumors. This anti-tumor activity is associated with increased caspase-3 activation and reduced tumor vascularity.
