LP12 + OVA8 + CpG + LLP2A is an experimental, programmable peptide nucleic acid (PNA)-based nanovaccine (PVN) designed for anticancer immune activation. The platform utilizes an 11-mer PNA scaffold (LP12) that facilitates the precise, "one-pot" assembly of multiple functional moieties via complementary PNA strand hybridization. This specific formulation incorporates three key components: an ovalbumin-derived antigenic peptide (OVA8, specifically the SIINFEKL epitope), a CpG oligonucleotide adjuvant, and an LLP2A ligand. The LLP2A ligand is designed to target activated α4β1 integrin (also known as VLA-4), which is expressed on both immune cells (such as dendritic cells) and certain tumor cells like melanoma. By targeting these cells, the vaccine enhances antigen presentation and stimulates a robust immune response involving CD8+ T cells and natural killer (NK) cells. In preclinical studies, this nanovaccine has demonstrated the ability to cause significant tumor regression and improve survival in syngeneic melanoma models.
