**LP143** is an oral, centrally acting, full agonist of the cannabinoid type 2 receptor (CB2), designed by Arena Pharmaceuticals and developed by Longboard Pharmaceuticals for central nervous system (CNS) diseases and disorders, particularly neurodegenerative conditions associated with neuroinflammation caused by microglial activation. It demonstrates over 1,000-fold selectivity for CB2 over CB1 in preclinical studies, minimizing psychoactive adverse effects, with sustained activity, favorable blood-brain-barrier penetration, and optimized pharmacology/pharmacokinetics compared to known compounds. Preclinical data in ALS mouse models showed delays in motor function loss and improved survival via regulation of neuroinflammatory processes; initial IND-enabling studies were completed in late 2021, but additional preclinical work was pursued before deciding on clinical advancement.[1][2][3][5][9]
