Overview

LPCN 1154 is an investigational oral formulation of brexanolone, a bioidentical version of the endogenous neuroactive steroid allopregnanolone. It is being developed for the rapid treatment of postpartum depression (PPD), aiming to provide a convenient, at-home alternative to the current standard of care, which requires intravenous administration and hospitalization. The drug acts as a positive allosteric modulator of the γ-aminobutyric acid (GABAA) receptor, enhancing inhibitory neurotransmission in the brain—a mechanism believed to underlie its antidepressant effects. LPCN 1154 has demonstrated bioequivalence with IV brexanolone in pivotal studies and is currently in Phase 3 clinical trials designed to support global registration for PPD treatment[2][3][5][10].
