LPD002 is a novel recombinant immunotherapeutic fusion protein consisting of a humanized anti-Programmed Cell Death Protein 1 (PD-1) monoclonal antibody linked to an engineered interleukin-2 (IL-2) mutein. Developed by Lepu Biopharma, the drug is designed to selectively deliver IL-2 signaling to PD-1-expressing tumor-infiltrating T cells, thereby enhancing anti-tumor effector functions while minimizing the systemic toxicities typically associated with high-dose IL-2 therapy. The IL-2 mutein component of LPD002 is engineered to retain binding activity to the high-affinity IL-2 receptor alpha (IL-2Rα/CD25) subunit but has attenuated affinity for the IL-2 receptor beta/gamma (IL-2Rβγ) complex. This receptor-biased design aims to promote the activation and proliferation of effector T cells within the tumor microenvironment while maintaining peripheral immune tolerance. Preclinical studies have demonstrated that LPD002 induces enhanced pSTAT5 signaling in PD-1+ cells and exhibits superior anti-tumor efficacy in both PD-1-sensitive and PD-1-resistant tumor models compared to PD-1 blockade alone.
