LPM3480392 (also known as LY03014) is a novel small molecule analgesic developed by Luye Pharma Group for the treatment of moderate to severe pain, including acute postoperative pain and breakthrough cancer pain. It acts as a G-protein-biased full agonist at the μ-opioid receptor (MOR), preferentially activating Gi signaling over β-arrestin-2 pathways. This bias is designed to provide potent analgesia with potentially fewer side effects compared to traditional opioids. Preclinical and early clinical studies have demonstrated its efficacy in animal models and healthy volunteers, with ongoing phase 2 trials in China for various pain indications[1][5][6][8].
