LPNP-p198 is a preclinical nanoparticle-based therapeutic designed to treat pancreatic ductal adenocarcinoma (PDAC), particularly cases resistant to gemcitabine. It consists of a polymeric nanoparticle carrier system (LPNP), composed of LGA-modified polyethyleneimine (PEI) and polylactic-co-glycolic acid (pLGA), which delivers a miR-198 mimic. The mechanism of action involves the downregulation of Valosin-Containing Protein (VCP) by miR-198, which disrupts autophagy maturation and sensitizes cancer cells to chemotherapy. In orthotopic mouse models, LPNP-p198 has demonstrated the ability to reduce tumor burden and metastases when used in combination with gemcitabine.
