LPT-99 is a small molecule drug that acts as an inhibitor of apoptotic protease-activating factor 1 (APAF1). It was initially developed by Loop Therapeutics and is being further developed by Spiral Therapeutics for the prevention and treatment of hearing loss, particularly to protect against cisplatin-induced ototoxicity. Preclinical studies have shown that LPT-99 can reduce apoptosis in cochlear cells exposed to cisplatin, thereby preserving auditory function. The drug has demonstrated dose-dependent protective effects on auditory function in animal models and improved cell survival in vitro. Its mechanism involves inhibition of APAF1, which plays a key role in the intrinsic pathway of apoptosis[1][3][4][5].
