Overview

LR26007 (also known as FMC-220) is an orally bioavailable small molecule designed to reactivate the p53 tumor suppressor protein in cancers harboring the Y220C mutation. The Y220C mutation is a common structural defect in the TP53 gene that creates a surface pocket, leading to protein destabilization and loss of function. LR26007 acts as a molecular chaperone, binding to this specific pocket to stabilize the mutant protein in its active, wild-type-like conformation. This restoration of p53 function triggers apoptosis and cell cycle arrest in tumor cells. Originally developed by First-in-Class (FMC) and licensed by LG Chem, the drug is currently undergoing Phase 1/2 clinical evaluation for the treatment of advanced solid tumors.
