LRE1 is a selective, small-molecule inhibitor of soluble adenylyl cyclase (sAC, also known as ADCY10), currently in preclinical development by Sacyl Pharmaceuticals as a non-hormonal male contraceptive. Discovered in 2016 as a research tool, LRE1 works by binding to the bicarbonate-binding site of sAC, thereby preventing the production of cyclic adenosine monophosphate (cAMP) in sperm. This inhibition reversibly impairs sperm motility and the ability to undergo capacitation, effectively preventing fertilization without affecting hormonal balance. While LRE1 serves as a foundational lead compound, Sacyl Pharmaceuticals is advancing optimized derivatives for clinical use.
