LRIG1 knockdown CD19 scFv CAR T cells are an experimental cellular immunotherapy developed by researchers at the Cleveland Clinic. These CAR T cells are engineered with a single-chain variable fragment (scFv) targeting the CD19 antigen, commonly found on B-cell malignancies, and are further modified to silence the expression of Leucine-Rich Repeats and Immunoglobulin-Like Domains 1 (LRIG1). LRIG1 has been identified as a novel T-cell checkpoint receptor that interacts with V-domain Immunoglobulin Suppressor of T cell Activation (VISTA) to inhibit T-cell signaling and limit the durability of CAR T cell responses. Preclinical studies in murine models of Raji lymphoma and melanoma have shown that LRIG1 ablation improves CAR T cell persistence and leads to long-term tumor regression, suggesting that targeting the LRIG1-VISTA axis may overcome common mechanisms of relapse in CAR T therapy.
