LRK-701 (also identified by the study code QSC301789) is an investigational, orally bioavailable small molecule inhibitor of Peptidyl-prolyl cis-trans isomerase NIMA-interacting 1 (Pin1), developed by Larkspur Biosciences. Pin1 is a unique regulatory enzyme that catalyzes the cis-trans isomerization of phosphorylated serine/threonine-proline motifs, acting as a molecular switch for numerous proteins involved in oncogenesis and immune evasion. By inhibiting Pin1, LRK-701 aims to disrupt multiple signaling pathways that tumors utilize to suppress the immune system, specifically by modulating the tumor microenvironment to reduce the activity of immunosuppressive myeloid cells and cancer-associated fibroblasts. The drug is currently being evaluated in Phase 1 clinical trials for patients with advanced solid tumors to assess its safety, pharmacokinetics, and preliminary therapeutic activity.
