Drug intelligence / Profile preview
LRP1-targeted PD-L1-blocking nanoconjugate (also known as P-12/AP-2/NCs) is an actively targeted immunotherapeutic polymeric nanoconjugate designed to cross the blood-brain barrier (BBB) and deliver PD-L1-blocking peptides to the tumor immune microenvironment (TiME). Developed by researchers at the Stanford University School of Medicine, the nanoconjugate is synthesized from a biodegradable, functionalized high molecular weight β-poly(l-malic acid) (PMLA) polymer backbone. This backbone is conjugated to two key peptide components: (1) Angiopep-2 (AP-2), which binds to low-density lipoprotein receptor-related protein 1 (LRP1) to facilitate receptor-mediated transcytosis across the BBB, and (2) P-12 (Aurigene NP-12), a small peptide antagonist that blocks the PD-1/PD-L1 and PD-1/PD-L2 pathways. The nanoconjugate is being investigated for the treatment of central nervous system malignancies, including glioblastoma multiforme (GBM) and melanoma brain metastases (MBM).
