LRP8-overexpression lentivirus is a gene therapy research tool designed to upregulate the expression of the Low-density lipoprotein receptor-related protein 8 (LRP8) gene. LRP8 is a member of the low-density lipoprotein receptor (LDLR) family and plays a critical role in cholesterol endocytosis. In multiple myeloma (MM), LRP8 is identified as an oncogenic factor that facilitates the uptake of cholesterol from the microenvironment, thereby activating the cholesterol-dependent mTORC1 signaling pathway. This activation promotes tumor cell proliferation, survival, and migration while suppressing autophagy. The lentiviral vector, typically constructed using a Ubi-MCS-3FLAG-CBh-gcGFP-IRES-puromycin backbone, is used in preclinical studies to establish stable cell lines and xenograft mouse models to investigate the metabolic reprogramming of cancer cells and evaluate LRP8 as a potential therapeutic target.
