LRRC15 antisense oligonucleotide is an experimental RNA-targeted therapy designed to silence the expression of Leucine-rich repeat-containing protein 15 (LRRC15). LRRC15 is a cell-surface protein and a well-known marker for myofibroblast-like cancer-associated fibroblasts (myCAFs), which contribute to the desmoplastic reaction, immunosuppression, and therapeutic resistance in solid tumors such as breast, pancreatic, and head and neck cancers. While LRRC15 is often associated with poor prognosis and is pursued as a therapeutic target in the tumor microenvironment, research presented by Haya Therapeutics indicates that direct knockdown of LRRC15 using ASOs may be insufficient to alter the disease-driving state of myCAFs or reduce tumor volume, suggesting that LRRC15 may be a downstream marker rather than a master regulator of the CAF phenotype.
