Overview

LRRC15-antiTGFβR2 is an investigational monoclonal antibody being developed by Bonum Therapeutics for the treatment of cancer. It is designed as a conditionally active therapeutic that targets both leucine-rich repeat-containing protein 15 (LRRC15) and transforming growth factor-beta receptor type 2 (TGFβR2). The antibody remains inactive in systemic circulation to prevent the toxicities associated with broad TGF-β inhibition. Upon reaching the tumor microenvironment, it binds to LRRC15, which is highly expressed on suppressive cancer-associated fibroblasts (CAFs). This binding triggers the activation of the anti-TGFβR2 component, allowing for localized inhibition of TGF-β signaling. This targeted approach aims to remodel the immunosuppressive stroma and enhance anti-tumor immune responses while minimizing systemic side effects.
