LRRC15-IFNα is a targeted cytokine therapy in preclinical development for cancer. It is designed as an engineered fusion protein that delivers interferon alpha (IFNα) activity specifically to cells expressing leucine-rich repeat-containing protein 15 (LRRC15), which is highly expressed on cancer-associated fibroblasts and certain tumor cells in the microenvironment of various solid tumors. The drug aims to activate immune responses locally within the tumor by binding to LRRC15, thereby avoiding the systemic toxicity associated with conventional IFNα therapies. Mechanistically, upon binding to LRRC15 on target cells, the IFNα moiety engages and activates type I interferon receptors (IFNAR), stimulating anti-tumor immunity while minimizing off-target effects. This approach leverages both targeted delivery and immune activation for potential therapeutic benefit in neoplasms[4][7][9].
