LSA-1 is a liver-stage antigen of Plasmodium falciparum that is expressed exclusively in infected hepatocytes during the preerythrocytic phase of malaria and is under development as a recombinant protein vaccine antigen. It is a large 230 kDa protein with a highly repetitive 17–amino acid central region flanked by conserved N- and C-terminal domains containing B- and T-cell epitopes, and is localized in the parasitophorous vacuole surrounding developing liver schizonts, where it is thought to contribute to liver schizogony and merozoite release.[1] Recombinant constructs of LSA-1, such as LSA-NRC (also known as falciparum malaria protein 011, FMP011), have been produced under GMP conditions and formulated with adjuvants (e.g., Montanide ISA 720) to induce strong humoral and cellular immune responses in animals and malaria-exposed humans, supporting its use as a preerythrocytic malaria vaccine candidate.[1]
