This is a multi-agent chemotherapy regimen composed of six drugs: LSA4, cyclophosphamide, methotrexate, daunomycin (also known as daunorubicin), L-asparaginase, and BCNU (carmustine). Such combinations are typically used in the treatment of hematologic malignancies like acute lymphoblastic leukemia (ALL) and certain lymphomas. Each component has a distinct mechanism of action: - Cyclophosphamide is an alkylating agent that crosslinks DNA to prevent cell division[1][2][3]. - Methotrexate is an antimetabolite that inhibits dihydrofolate reductase, blocking DNA synthesis. - Daunomycin/daunorubicin is an anthracycline antibiotic that intercalates into DNA and inhibits topoisomerase II. - L-asparaginase depletes asparagine from plasma, starving leukemic cells unable to synthesize this amino acid[6][8][10]. - BCNU (carmustine) is a nitrosourea alkylating agent causing DNA crosslinking. The combination targets rapidly dividing cancer cells through multiple cytotoxic mechanisms.
