Overview

LSN2148936 (also known as VDRM2) is a nonsecosteroidal small molecule agonist of the Vitamin D Receptor (VDR). Developed by Eli Lilly, it was designed to overcome the dose-limiting hypercalcemia associated with traditional secosteroidal VDR ligands like calcitriol. In preclinical studies, LSN2148936 demonstrated the ability to restore bone mineral density in osteopenic models with a significantly wider therapeutic window (57-fold) compared to natural ligands. It has been investigated for potential applications in osteoporosis and prostate cancer, and serves as a structural reference for the development of photopharmacological VDR agonists for skin conditions like psoriasis.
