Overview

The LspA inhibitor is a preclinical-stage small molecule antibiotic being developed by Prokaryotics, Inc. It targets lipoprotein signal peptidase II (LspA), an essential bacterial enzyme responsible for the maturation of lipoproteins and the maintenance of cell envelope integrity. By inhibiting LspA, the compound disrupts the processing and assembly of lipoproteins, which compromises the bacterial cell envelope and leads to growth inhibition or cell death. This target is highly conserved across various bacterial species, making it a promising candidate for the treatment of multidrug-resistant (MDR) bacterial infections, particularly those caused by Gram-negative pathogens. The program is supported by NIH/NIAID funding.
