Overview

LTβR agonist monoclonal antibodies are a class of immunotherapeutic biologics designed to target and activate the lymphotoxin-β receptor (LTβR), a member of the tumor necrosis factor receptor (TNFR) superfamily. Activation of LTβR signaling is essential for the development of secondary lymphoid organs and the induction of tertiary lymphoid structures (TLS) within the tumor microenvironment. By promoting the formation of high endothelial venules (HEVs) and enhancing the recruitment of T cells and dendritic cells, these antibodies aim to convert 'cold' tumors into 'hot' tumors, thereby improving anti-tumor immune responses. Several specific agents have been explored, including Pfizer's tetravalent antibody PF-07329640, Roche's RO7567132, and Mestag Therapeutics' FAP-conditional agonist MST-0300. While early research (e.g., CBE11) demonstrated potent anti-tumor activity in preclinical models, clinical development remains in early stages, with some programs facing strategic terminations.
