LT-025 is a homogeneous antibody-drug conjugate (ADC) specifically designed for the treatment of renal cell carcinoma (RCC). It targets the kidney injury molecule-1 (KIM-1) receptor, which is highly expressed on RCC cells but has limited expression in healthy tissues. The ADC is synthesized using a site-specific enzymatic conjugation technology involving microbial transglutaminase (MTGase), which attaches the cytotoxic payload DM1 (mertansine), a potent microtubule inhibitor, to the Fc region of a KIM-1-specific antibody. This conjugation method yields a well-defined drug-to-antibody ratio (DAR) of 2. Preclinical studies have shown that LT-025 demonstrates superior cytotoxicity against metastatic and sunitinib-resistant RCC models compared to standard treatments, and it exhibits an improved safety profile due to its homogeneous nature.
