LT-1951 is a proprietary topical formulation of the BRAF inhibitor encorafenib, developed by Lutris Pharma. It is specifically designed to treat skin toxicities, such as acneiform rash, induced by Epidermal Growth Factor Receptor (EGFR) inhibitors or MEK inhibitors used in cancer therapy. The drug operates through a mechanism known as 'paradoxical activation' of the mitogen-activated protein kinase (MAPK) pathway. In BRAF wild-type cells, such as keratinocytes, the inhibition of BRAF leads to the activation of downstream signaling, which promotes cell proliferation and survival. By locally applying LT-1951 to the skin, this paradoxical effect counteracts the inhibitory signals from systemic oncology treatments, promoting skin healing and reducing disfiguring rashes without interfering with the systemic anti-tumor efficacy of the primary cancer therapy.
