LTBR-armored gamma-delta CAR T cells are an investigational allogeneic cell therapy consisting of gamma-delta (γδ) T cells engineered to express a chimeric antigen receptor (CAR) targeting Claudin 6 (CLDN6) and synthetically reprogrammed with the Lymphotoxin beta receptor (LTβR). Developed by Arsenal Bio, this therapy utilizes a genome-scale discovery approach to overcome the immunosuppressive tumor microenvironment (TME). The overexpression of LTβR, a TNF superfamily receptor not typically found in T cells, confers resistance to inhibitory signals such as TGF-β, adenosine, and regulatory T cells (Tregs) while promoting an αβ-like effector program. This results in enhanced cytotoxicity, increased persistence, and improved metabolic fitness. In preclinical models of aggressive ovarian cancer, these armored cells have demonstrated the ability to mediate complete and durable tumor regressions, offering a potential off-the-shelf solution for solid malignancies.
