Overview

LTCR-N1-1 is an autologous T-cell receptor engineered T-cell (TCR-T) therapy developed by Lion TCR for the treatment of Epstein-Barr Virus (EBV)-associated malignancies, such as nasopharyngeal carcinoma. The therapy utilizes the LioCyx-M platform, which involves isolating a patient's own T cells and using mRNA electroporation to transiently express TCRs specific to EBV antigens. This approach allows the engineered T cells to recognize and eliminate EBV-infected tumor cells. The use of mRNA transfection provides a safety advantage by ensuring transient expression of the TCR, which may reduce the risk of long-term off-target toxicities compared to permanent viral vector-based modifications. LTCR-N1-1 is currently being evaluated in early-phase clinical trials for EBV-related cancers.
