Overview

LTG-321 is an investigational small molecule developed by Latigo Biotherapeutics as a selective modulator of the sodium channel protein Nav1.8 (Sodium channel protein type 8 subunit alpha). It is being developed as an oral, non-opioid treatment for chronic pain, aiming to block pain signals at the level of peripheral sensory neurons and thus prevent their transmission to the central nervous system. LTG-321 is a follow-on compound to Latigo's earlier pain candidate (LTG-001), reportedly featuring unique chemical scaffolding that may provide improved potency for chronic pain indications. As of September 2025, it is in Phase 1 clinical studies in healthy adults for safety, tolerability, pharmacokinetics, pharmacodynamics, and pain tolerance using cold pressor assays.
