Overview

LTI-03 is a synthetic seven-amino acid peptide derived from the caveolin scaffolding domain (CSD) of the caveolin‑1 (Cav1) protein. It is designed as an inhaled therapy for idiopathic pulmonary fibrosis (IPF). The drug acts by restoring the balance of biological signals in the lung, inhibiting pro-fibrotic signaling pathways, and promoting survival and function of alveolar epithelial cells. Cav1 normally prevents fibrosis by maintaining a balance between pathways that initiate and arrest lung repair; its expression is reduced in IPF lungs. By mimicking Cav1 activity, LTI‑03 aims to slow or reverse lung function decline through both anti-fibrotic effects and protection of healthy epithelial cells[5][6][8]. The drug has shown positive trends in biomarker reduction related to fibrosis and inflammation in early clinical trials[6][7].
