Overview

LTI-05 is a novel small molecule drug candidate that acts as an epithelial sodium channel (ENaC) inhibitor. It is being developed primarily as an antifibrotic agent, with its main indication currently in preclinical development for cystic fibrosis. The drug was originally developed by Lung Therapeutics and is now under the development of Rein Therapeutics (formerly Aileron Therapeutics). As an ENaC antagonist, LTI-05 targets the epithelial sodium channel to reduce sodium absorption in airway epithelia, which may help improve airway hydration and mucociliary clearance—key therapeutic goals in cystic fibrosis management. LTI-05 has not received orphan drug designation and remains a new molecular entity[1][2].
