LTI-2 is an optimized lipid nanoparticle (LNP) formulation designed for the high-efficiency delivery of mRNA payloads into resting primary human T-cells. Developed by Lentigen Technology Inc. (a Miltenyi Biotec company), LTI-2 was identified through a design of experiment (DOE) approach using a library of ionizable lipids. Unlike conventional LNPs used in commercial vaccine formulations, LTI-2 achieves robust transfection without the need for exogenous T-cell activation or ApoE protein supplementation, utilizing an uptake mechanism independent of the low-density lipoprotein receptor (LDLR). This formulation is intended to streamline the production of chimeric antigen receptor (CAR) T-cell therapies for indications such as leukemia, lymphoma, and autoimmune diseases, potentially enabling both ex-vivo and in-vivo engineering by overcoming the requirement for strong activation and specific protein supplementation.
