LTX-04 is an immediate-release oral formulation of hydromorphone developed by Acura Pharmaceuticals using its proprietary LIMITx technology. The primary goal of this formulation is to deter abuse by limiting the rate and extent of hydromorphone release when multiple tablets are ingested. LIMITx technology works by incorporating buffering ingredients that neutralize stomach acid as more tablets are consumed, thereby retarding drug release and reducing peak blood concentrations (Cmax) in overdose or abuse scenarios. Clinical studies demonstrated that a single tablet achieves therapeutic blood levels but with lower Cmax compared to marketed hydromorphone products; when multiple tablets are taken, Cmax is further reduced—up to 65% less than comparators—potentially lowering the risk of overdose from excessive oral intake. The product was granted FDA Fast Track designation for its potential to address opioid abuse[1][2][3][4][5][6][8].
