Overview

LTX-401 is a novel oncolytic amino acid derivative developed by Lytix Biopharma designed for the local treatment of solid tumors. It acts as a cytolytic immunotherapeutic agent, primarily administered via intratumoral injection. The compound exhibits membranolytic activity, causing rapid necrotic cell death in cancer cells and leading to the release of damage-associated molecular patterns (DAMPs) such as calreticulin, ATP, and HMGB1. This process promotes immunogenic cell death and stimulates antitumor immune responses. Mechanistically, LTX-401 accumulates in the Golgi apparatus of tumor cells and induces permeabilization of intracellular organelles including mitochondria and lysosomes. Preclinical studies have demonstrated its efficacy in inducing tumor regression and long-term protective effects in animal models of melanoma, fibrosarcoma, and hepatocellular carcinoma. The drug is being developed for use against deep-seated solid tumors.
