Lu AF34134 is a small molecule antagonist of the G protein-coupled receptor 6 (GPR6), developed by Lundbeck for the treatment of Parkinson's disease. GPR6 is an orphan G protein-coupled receptor that is highly and specifically expressed in the striatopallidal medium spiny neurons of the indirect pathway within the basal ganglia. In Parkinson's disease, the indirect pathway is overactive, contributing to motor deficits; by blocking GPR6, Lu AF34134 aims to reduce this overactivity and restore motor function. The drug has been evaluated in Phase 1 clinical trials to assess its safety, tolerability, and pharmacokinetics, including a food interaction crossover study in healthy young and elderly volunteers.
