Lu AG13909 is a humanized monoclonal antibody that targets and neutralizes adrenocorticotropic hormone (ACTH) by binding to it with high affinity. By blocking ACTH from binding to the melanocortin 2 receptor in the adrenal glands, Lu AG13909 inhibits ACTH-induced signaling, resulting in decreased secretion of glucocorticoids (such as cortisol), mineralocorticoids (such as aldosterone), and androgens. This mechanism addresses conditions characterized by excessive ACTH activity, such as Cushing's disease and classic congenital adrenal hyperplasia (CAH). The drug is being developed by Lundbeck and is currently undergoing Phase 2 clinical trials for Cushing's disease and Phase 1 trials for CAH. Preclinical studies have shown significant reductions in corticosterone/cortisol levels without notable adverse effects[1][2][3][5].
