Lu AG22515 is a recombinant fusion antibody designed as a bispecific molecule targeting CD40 ligand (CD40L) and human serum albumin. It consists of anti-CD40L single-chain variable fragments (scFvs) fused to an anti-human serum albumin Fab fragment. The drug acts as a CD40 ligand inhibitor and is being developed primarily for the treatment of thyroid eye disease (TED), also known as Graves ophthalmopathy—an autoimmune inflammatory disorder associated with Graves' disease that causes inflammation and swelling behind the eyes leading to proptosis (eye bulging). The mechanism involves modulation of immune responses by inhibiting the interaction between CD40L and its receptor, which plays a key role in autoimmune pathogenesis. Lu AG22515 is currently in Phase 1 clinical trials for TED/Graves ophthalmopathy[3][4][5].
