Drug intelligence / Profile preview
Lu-Tamrada (also known as 177Lu-Tamrada) is a first-in-class nanomedicine radiopharmaceutical developed by Ironfist Therapeutics (a sister company of Programmable Medicine) for the treatment of solid tumors. It consists of a hydroxyl polyamidoamine (PAMAM) dendrimer nanomedicine conjugated to a DOTA chelator that captures the beta-emitting radioisotope lutetium-177 (177Lu). Unlike conventional radiopharmaceuticals that target tumor-specific antigens, Lu-Tamrada is tumor-agnostic and selectively targets tumor-associated macrophages (TAMs) in the tumor microenvironment without requiring a targeting ligand. Upon uptake by TAMs via receptor-independent endocytosis, the drug is retained intracellularly, delivering localized radiation that ablates immunosuppressive TAMs, halts tumor growth, and re-sensitizes the tumor microenvironment to immune checkpoint inhibitors.
