Lu-TLX250-TRAP is a multi-functional radiopharmaceutical therapy (RPT) candidate developed by Telix Pharmaceuticals for the treatment of triple-negative breast cancer (TNBC). It is a fusion protein consisting of an anti-carbonic anhydrase IX (CAIX) antibody fused to the ectodomain of the TGFβ receptor II (TGFβRIIect), labeled with the beta-emitting radioisotope lutetium-177 (Lu-177). The agent is designed to localize to CAIX-expressing tumor cells, delivering cytotoxic radiation while simultaneously sequestering and neutralizing TGFβ in the tumor microenvironment. This dual mechanism aims to counteract TGFβ-mediated immunosuppression and radioresistance, thereby enhancing the recruitment and activity of CD4+ and CD8+ T cells and improving overall antitumor efficacy.
