LUA004 is a novel antibody-drug conjugate (ADC) targeting trophoblast glycoprotein (5T4), an oncofetal protein frequently upregulated in various solid tumors, including esophageal, head-and-neck, and lung cancers. Developed by Qilu Pharmaceutical, LUA004 utilizes a site-specific conjugation technology to achieve a drug-to-antibody ratio (DAR) of 8. It incorporates a proprietary cleavable linker and a novel microtubule inhibitor payload, QLS8152. Preclinical studies demonstrate that LUA004 exhibits sub-nanomolar binding affinity, potent in vitro cytotoxicity, and robust dose-dependent tumor inhibition in xenograft models. Notably, LUA004 has shown an improved therapeutic window and superior tolerability compared to traditional vedotin-based ADCs in non-human primate studies, indicating a significant increase in the therapeutic index.
