LUA006 is a preclinical-stage bispecific antibody-drug conjugate (ADC) targeting both Epidermal Growth Factor Receptor (EGFR) and B7-H3 (CD276). Developed by QiLu Pharmaceutical, the agent is designed with attenuated EGFR binding to minimize on-target, off-tumor toxicities in normal tissues while maintaining enhanced B7-H3 binding and internalization dynamics to improve anti-tumor efficacy. Uniquely, LUA006 incorporates two distinct payload classes with complementary mechanisms of action and sequential release kinetics to address tumor heterogeneity and overcome payload-associated resistance. It has demonstrated potent activity in various solid tumor models, including non-small cell lung cancer (NSCLC), esophageal squamous cell carcinoma (ESCC), and head and neck squamous cell carcinoma (HNSCC).
