LUA011 is a novel bispecific antibody-drug conjugate (ADC) targeting both Cadherin-17 (CDH17) and the Hepatocyte Growth Factor Receptor (c-Met), developed by Qilu Pharmaceutical. It is designed for the treatment of gastrointestinal (GI) malignancies, including colorectal, gastric, and pancreatic cancers, where these two targets are frequently co-expressed. The ADC utilizes a proprietary topoisomerase I inhibitor (TOPO1i) payload conjugated to the bispecific antibody via a glycan-based platform. By dual-targeting CDH17 and c-Met, LUA011 aims to improve the therapeutic index and mitigate the on-target gastrointestinal toxicities often observed with mono-specific CDH17 ADCs, as the co-expression of these targets is rare in normal tissues. Preclinical data presented at AACR 2026 demonstrated potent anti-tumor activity, rapid internalization, and a favorable safety profile in animal models, with a maximum tolerated dose exceeding 30 mg/kg in cynomolgus monkeys.
