lucansartuzumab

Lucansartuzumab is a TROP2-directed antibody-drug conjugate (ADC) currently under clinical investigation for the treatment of non-small cell lung cancer (NSCLC). The drug is composed of a humanized monoclonal antibody targeting the trophoblast cell-surface antigen 2 (TROP2), which is frequently overexpressed in various epithelial tumors, including NSCLC. The antibody is conjugated to a cytotoxic payload, which has been described in trial contexts as lurbinectedin (or a derivative thereof), a DNA minor groove binder that inhibits activated transcription and induces double-strand breaks. Lucansartuzumab is being evaluated in combination with the PD-1 inhibitor tislelizumab as a neoadjuvant therapy for patients with resectable stage II–IIIB NSCLC to improve pathological complete response (pCR) rates and overall survival outcomes.
