LUCAR-DKS1 is a chimeric antigen receptor (CAR) natural killer (NK) cell therapy being developed for the treatment of relapsed or refractory autoimmune diseases[1][2][4][6]. The therapy involves engineering NK cells to express a CAR that targets disease-relevant antigens, enabling directed immune activity against pathogenic cells implicated in various autoimmune conditions. It is primarily developed by Legend Biotech, and clinical trials are currently evaluating its safety, tolerability, pharmacokinetics, and efficacy in adult patients with relapsed/refractory autoimmune diseases including systemic lupus erythematosus (SLE), systemic sclerosis (SSc), ANCA-associated vasculitis (AAV), idiopathic inflammatory myopathies (IIM), and Sjögren's syndrome (SjS)[2][4][6]. This therapy represents a novel, cell-based immunotherapy approach aimed at providing disease-modifying effects in severe autoimmunity.
