Overview

Lucatumumab is a fully human monoclonal antibody that targets the CD40 antigen, a member of the tumor necrosis factor receptor superfamily, expressed on B cells and various malignancies. It acts as an antagonist by binding to CD40 and blocking its interaction with CD40 ligand (CD40L), thereby inhibiting CD40L-induced cell proliferation and survival signals in malignant B cells. Lucatumumab also mediates antibody-dependent cellular cytotoxicity (ADCC), leading to immune-mediated lysis of tumor cells overexpressing CD40. Developed initially by Chiron using Abgenix's XenoMouse technology, it was later advanced by Novartis after acquiring Chiron, in collaboration with XOMA. The drug was investigated for hematological malignancies such as multiple myeloma, chronic lymphocytic leukemia (CLL), follicular lymphoma, non-Hodgkin lymphoma, and Hodgkin lymphoma but development was discontinued after Phase 1/2 trials due to limited efficacy[1][2][3][4][5][6].
