Overview

lucitanib + toripalimab is an investigational combination therapy comprising a multi-target tyrosine kinase inhibitor and a PD-1 blocking monoclonal antibody. **Lucitanib** (also known as AL3810) is a potent oral inhibitor of vascular endothelial growth factor receptors (VEGFR1, VEGFR2, VEGFR3), fibroblast growth factor receptors (FGFR1, FGFR2), and platelet-derived growth factor receptors (PDGFRα, PDGFRβ). By targeting these pathways, lucitanib exerts anti-angiogenic and anti-proliferative effects on tumor cells. **Toripalimab** is a humanized IgG4 monoclonal antibody that acts as an immune checkpoint inhibitor by binding to the programmed cell death protein 1 (PD-1) receptor on T cells. This blockade prevents the interaction between PD-1 and its ligands (PD-L1 and PD-L2), thereby restoring and enhancing the anti-tumor immune response. The combination is currently being evaluated in a Phase II clinical trial (NCT05616533) sponsored by Sun Yat-sen University for the treatment of patients with PD-L1-positive, HR-positive/HER2-negative advanced breast cancer who have progressed on prior CDK4/6 inhibitor therapy.
